OBJECTIVE: To develop a quantitative reversed phase high performance liquid chromatography method using diode array detector for the determination of ursolic acid and lupeol in leaves of Ilex cornuta. METHODS: An Agilent Zorbax ODS column was used with a gradient elution composed of acetonitrile and 0.1% phosphoric acid solution at a flow rate of 1.0 mL-min(-1), Shionone was used as the internal standard. RESULTS: This method was validated in term of linearity, accuracy, precision, reproducibility and stability. The method was applied to analyze leaves of Ilex cornuta from 10 different locations. As a result, the contents of ursolic acid and lupeol were 1.23%-1.63% and 0.59%-0.95%, respectively. CONCLUSION: The established method was proved to be simple, reproducible, accurate, and can be used to evaluate the quality of leaves of Ilex cornuta.
